小鼠组织因子途径抑制物〖TFPI〗elisa试剂盒利用抗原抗体之间专一性键结之特性，对检体进行检测；由于结合于固体承载物(一般为塑胶孔盘)上之抗原或抗体仍可具有免疫活性，因此设计其键结机制后，配合酵素呈色反应，即可显示特定抗原或抗体是否存在，并可利用呈色之深浅进行定量分析。根据待测样品与键结机制的不同，ELISA可设计出各种不同类型的检测方式，主要以夹心法（sandwich）、间接法（indirect）、以及竞争法（Competitive）三种为主。

在ELISA中，进行各项实验条件的选择是很重要的，其中包括：
（1）固相载体的选择：许多物质可作为固相载体，如聚氯乙烯、聚苯乙烯、聚丙酰胺和纤维素等。其形式可以是凹孔平板、试管、珠粒等。目前常用的是40孔聚苯乙烯凹孔板。不管何种载体，在使用前均可进行筛选：用等量抗原包被，在同一实验条件下进行反应，观察其显色反应是否均一性，据此判明其吸附性能是否良好。
（2）包被抗体(或抗原)的选择：将抗体(或抗原)吸附在固相载体表面时，要求纯度要好，吸附时一般要求PH在9.0～9.6之间。吸附温度，时间及其蛋白量也有一定影响，一般多采用4℃18～24小时。蛋白质包被的zui适浓度需进行滴定：即用不同的蛋白质浓度(0.1、1.0和10μg/ml等)进行包被后，在其它试验条件相同时，观察阳性标本的OD值。选择OD值zui大而蛋白量zui少的浓度。对于多数蛋白质来说通常为1～10μg/ml。
（3）酶标记抗体工作浓度的选择：首先用直接ELISA法进行初步效价的滴定(见酶标记抗体部份)。然后再固定其它条件或采取“方阵法”(包被物、待检样品的参考品及酶标记抗体分别为不同的稀释度)在正式实验系统里准确地滴定其工作浓度。 
（4）酶的底物及供氢体的选择：对供氢体的选择要求是价廉、安全、有明显地显色反应，而本身无色。有些供氢体(如OPD等)有潜在的致癌作用，应注意防护。有条件者应使用不致癌、灵敏度高的供氢体，如TMB和ABTS是目前较为满意的供氢体。底物作用一段时间后，应加入强酸或强碱以终止反应。通常底物作用时间，以10-30分钟为宜。底物使用液必须新鲜配制，尤其是H2O2在临用前加入。
