本试剂盒应用双抗体夹心法测定标本中鸡分泌型免疫球蛋白A(sIgA)测定试剂盒水平。用纯化的抗体包被微孔板，制成固相抗体，往包被单抗的微孔中依次加入，再与HRP标记的结合，形成抗体-抗原-酶标抗体复合物，经过*洗涤后加底物TMB显色。TMB在HRP酶的催化下转化成蓝色，并在酸的作用下转化成zui终的黄色。颜色的深浅和样品中的呈正相关。用酶标仪在450nm波长下测定吸光度（OD值），通过标准曲线计算样品中的浓度。

加样在ELISA中一般有3次加样步聚，即加标本，加酶结合物，加底物。加样时应将所加物加在LEISA板孔的底部，避免加在孔壁上部，并注意不可溅出，不可产生气泡。加标本一般用微量加样器，按规定的量加入板孔中。每次加标本应更换吸嘴，以免发生交叉污染，也可用一次性的定量塑料管加样。有此测定（如间接法ELISA）需用稀释的血清，可在试管中按规定的稀释度稀释后再加样。也可在板孔中加入稀释液，再在其中加入血清标本，然后在微型震荡器上震荡1分钟以保证混和。加酶结合物应用液和底物应用液时可用定量多道加液器，使加液过程迅速完成。
